Methylene blue has been investigated as a potential treatment for neurodegenerative diseases, including Alzheimer’s disease, due to its neuroprotective, antioxidant properties and its ability to influence the mechanisms underlying the disease.
How Methylene Blue Works in Alzheimer’s
- Inhibition of Tau Protein Aggregation:
- In Alzheimer’s, abnormal tau proteins form tangles (neurofibrillary tangles) that damage brain cells.
- Methylene blue has shown the ability to inhibit the aggregation of tau proteins and even dissolve existing tangles, helping to protect neurons.
- Reduction of Oxidative Stress:
- It acts as an antioxidant, neutralizing free radicals that contribute to cell damage and cognitive decline in Alzheimer’s disease.
- Improvement in Mitochondrial Function:
- Methylene blue can act as an electron carrier in mitochondria, enhancing energy (ATP) production. This is critical because brain cells in Alzheimer’s often suffer from mitochondrial dysfunction.
- Neuroprotection and Synaptic Plasticity:
- It supports the ability of neurons to form new connections (synaptic plasticity), which is essential for learning and memory.
Research Evidence
A stabilized form of methylene blue, known as LMTM (Leuco-Methylthioninium Mesylate), has been tested in human clinical trials. Some studies reported positive effects in slowing cognitive decline in patients with mild to moderate Alzheimer’s.
Preclinical Studies: Animal models of Alzheimer’s have shown improvements in memory and reductions in Alzheimer’s-related pathology, such as tau tangles, with methylene blue treatment.
